同时BD Clontech还提供对照质粒,包括pSIREN-Shuttle、pSIREN-DNR、pSIREN-RetroQ三种载体的萤光素酶阳性对照和阴性对照。
BD clontech公司新推出的RNAi-Ready pSIREN-RetroQ-ZsGreen 和 pSIREN-DNR-DsRedExpress是带有荧光蛋白基因的即用型RNAi载体。这些荧光蛋白(分别为绿色荧光和红色荧光)不需要加任何辅助因子或底物就可以在许多细胞系中表达。由于这些蛋白发出的荧光非常强,可以用荧光显微镜或者是流式细胞仪来观察。不但可以直接观察转染的效果,还可以收集阳性转染细胞。同时荧光蛋白的表达和shRNA的表达时独立的,因此不影响shRNA基因沉默的效果。RNAi-Ready pSIREN-RetroQ-ZsGreen可以作为质粒操作,也可以通过相应的细胞系包装成重组逆转录病毒;pSIREN-DNR-DsRedExpress Vectors则可以通过Knockout Adenoviral RNAi System 2来包装成重组腺病毒。(见figure 8)
Figure 8. Fluorescent RNAi-Ready pSIREN vectors generate effective, tagged shRNA expression cassettes (SECs). Using the BDTM Knockout RNAi Clone & Confirm PCR Kit, fluorescence-tagged SECs were generated by PCR from ligation mixtures of a negative control (–) or a luciferase (+) shRNA annealed oligo, and an RNAi-Ready pSIREN vector with a fluorescent marker. The PCR-generated SECs were subsequently cotransfected with pCMV-Luc into HEK 293 cells. Luciferase activity was measured 48 hours after transfection. Shown are ZsGreen-tagged (Panel A) and DsRedExpress-tagged (Panel B) SECs in cotrans-fected cells. The SECs effectively knock down luciferase expression by >85% (Panel C).
