User Bulletin #2 ABI P RISM 7700 Sequence Detection System
IntroductionAmplification of an endogenous control may be performed to standardize the amount of sample RNA or DNA added to a reaction. For the quantitation of gene expression, researchers have used ß-actin, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), ribosomal RNA (rRNA), or other RNAs as this endogenous control. Relative quantitation with data from the ABI P RISM® 7700 Sequence Detection System (using version 1.6 software) can be performed using the standard curve method or the comparative method. The availability of distinguishable reporter dyes for the ABI P RISM 7700 Sequence Detection System makes it possible to amplify and detect the target amplicon and the endogenous control amplicon in the same tube (multiplex polymerase chain reaction [PCR]).
