Commercial success of approved biopharmaceuticals and growing numbers of protein-based drug candidates have led to projections of metric tons of monoclonal antibodies (MAbs) being required in a few short years. To meet this demand cell culture capacity is i with reactors in the 12-15 000 L range and larger coming on-line. Expression levels, currently in the 0.1-1.0 g/L range, are expected to increase several-fold. In downstream processing, efforts are directed at improving process economy by decreasing the number and cost of unit operations. Current trends in antibody production indicate an increased use of protein A chromatography media for product capture.
